Notes Perform all handling and transcription reactions in an RNase-free environment. Wearing gloves and adding RNase inhibitors to the reaction are recommended to prevent RNA degradation.
For transcripts shorter than 100 nt, the template amount can be increased up to 2 µg.
To avoid precipitation caused by DNA and spermidine interaction at low temperatures, prepare the reaction mix at room temperature.
