Description Dpn I is a restriction endonuclease that specifically recognizes and cleaves methylated or hemimethylated adenine residues within the sequence 5′-Gm6ATC-3′. It is widely used in molecular cloning and site-directed mutagenesis to selectively digest parental DNA while leaving newly synthesized DNA intact. Due to its high specificity for methylated DNA, Dpn I is a critical tool in plasmid preparation, mutation screening, and DNA replication studies in both research and diagnostic assay development.
