E. coli Single-Strand DNA Binding Protein (SSB) is essential for DNA replication and repair. Forms tetramers that specifically bind 8–16 nucleotides, protecting ssDNA from nuclease degradation. In vitro, SSB stabilizes ssDNA regions and relaxes dsDNA, enhancing polymerase activity.
